Chromatographic Studies on Nucleic Acids

چکیده

The nucleic acids are such universal constituents of biological systems and their function is probably so important that it is not surprising that much labour has been spent on the study of their composition. Until very recently there have been no methods of analysis which could be considered more than semiquantitative, and it now seems doubtful whether many published analyses are even approximate. While it is now possible to separate and estimate quantitatively mixtures of purine and pyrimidine bases and some of their derivatives by means of paper strip chromatography (Hotchkiss, 1948; Vischer & Chargaff, 1948; Markham & Smith, 1949b), it is by no means as simple to convert nucleic acids into these compounds quantitatively. We have investigated many methods for the hydrolysis and estimation of nucleic acids, and have found it impossible to get satisfactory results by any published method. By combining our chromatographic technique with a simple method of quantitative hydrolysis and a solvent system which resolves the hydrolysate, we have evolved a new method of analysis which, in addition to being more sensitive than existing methods, is accurate and simple.

Chromatographic studies of nucleic acids. 4. The nucleic acid of the turnip yellow mosaic virus including a note on the nucleic acid of the tomato bushy stunt virus.

It was largely because of our interest in the turnip yellow mosaic virus (Markham & Smith, K. M., 1949) that we began our study of nucleic acids. This virus contains about 37 % by weight of ribonucleic acid, which is an unusually high proportion. All the infected plants which we have examined contain, as well as the virus, another characteristic and related protein in an amount equivalent to 25...

A chromatographic comparison of the nucleic acids from isologous newborn, adult, and neoplastic thymus.

The total tissue nucleic acids (DNA and RNA) have been isolated from normal and neoplastic mouse thymus glands and chromatographically fractionated. Significant differences in the chromatographic distribution of the nucleic acids from normal adult thymus and x-radiation-induced thymic lymphosarcoma were found. Chromatographic patterns intermediate between normal and tumor were found in the thym...

A Chromatographic Study of Hydrolysis in the Feulgen Nucleal Reaction* by Philip

Acid hydrolysis of tissue has long been recognized as a necessary part of the Feulgen method for demonstrating the presence of DNA (deoxyribonucleic acid) in histological preparations (Feulgen and Rossenbeck, 1924). When attempts were made to use the method for the quantitative determination of DNA (Pollister and Ris, 1947; Di Stefano, 1948; Ris and Mirsky, 1949; Swift, 1950 a), the role of hyd...

Purine and Pyrii~idlne Contents of Some Desoxypentose N,ucleic Acids by M. M. Daly,* v. G. Allfrey, Ai~ A. E. Mirsky

In this paper the pyrimidine and purine contents of nucleic acids of the desoxypentose type are given. The nucleic acids analyzed were prepared from different tissues of the same organism and from a variety of different organisms. Preparations from eleven different sources were studied. In many of the preparations it was possible to account for all of the nucleic acid phosphorus present; and in...
